The First input is a combo input ( 1/4" or XLR) -  this means that it can accept either a ¼”  Jack ( like a guitar or keyboard cable), Or it can accept a microphone (XLR cable). The Combo input gets independent Reverb effect to make your voice ( or instrument ) sound just the way you want. 

The Second input is a 1/4" inch only - which means you can plug in either a guitar or keyboard or any other instrument. This second input has an independant reverb and chorus effect, and also a 3-Band EQ ( High Mid Low)

The D.I out can be used in live scenarios, where you want to set your ideal sound on the amp and send that output to the sound system at the venue with exactly the sounds and effects that you want to be heard outside.
